NM_002519.3(NPAT):c.422C>T (p.Pro141Leu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P141L variant (also known as c.422C>T), located in coding exon 6 of the NPAT gene, results from a C to T substitution at nucleotide position 422. The proline at codon 141 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
