NM_000492.4(CFTR):c.472A>T (p.Ser158Cys) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S158C variant (also known as c.472A>T), located in coding exon 4 of the CFTR gene, results from an A to T substitution at nucleotide position 472. The serine at codon 158 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
