NM_000384.3(APOB):c.9667T>C (p.Tyr3223His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y3223H variant (also known as c.9667T>C), located in coding exon 26 of the APOB gene, results from a T to C substitution at nucleotide position 9667. The tyrosine at codon 3223 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
