Pathogenic for Neuromuscular disease caused by qualitative or quantitative defects of dystrophin — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000023.10:g.(31893491_31947712)_(31950345_31986455)del, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the deletion of exons 46-47 in the DMD gene. A presumed nomenclature of c.(6614+1_6615-1)_(6912+1_6913-1)del has been designated for the purposes of this classification. Although exact breakpoints of this deletion are not known, it is expected to result in a frameshift in the DMD gene, a known mechanism of disease. The variant was absent in 16115 control chromosomes in the gnomAD database (structural variants dataset). c.(6614+1_6615-1)_(6912+1_6913-1)del has been reported in the literature in several individuals affected with Duchenne Muscular Dystrophy (e.g. Onegut_2000, Carsana_2005, del Gaudio_2008, Nallamilli_2021). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three submitters have provided clinical-significance assessments for this (or similar) variants to ClinVar after 2014, and classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic.
