Likely pathogenic for Polycystic kidney disease 2 — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000297.4(PKD2):c.1095-1G>T, citing LabCorp Variant Classification Summary - May 2015: Variant summary: PKD2 c.1095-1G>T is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. One in silico splicing tool predicts the variant to be possibly pathogenic (TraP). However, this prediction has not yet been confirmed by functional studies. The variant was absent in 247946 control chromosomes (gnomAD). To our knowledge, no occurrence of c.1095-1G>T in individuals affected with Polycystic Kidney Disease 2 and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
