NM_006231.4(POLE):c.4519_4520del (p.Gln1507fs) was classified as Uncertain significance by Genetic Services Laboratory, University of Chicago. This variant lies in the POLE gene (transcript NM_006231.4) at coding-DNA position 4519 through coding-DNA position 4520, deleting 2 bases; at the protein level this means shifts the reading frame starting at glutamine residue 1507, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: DNA sequence analysis of the POLE gene demonstrated a two base pair deletion in exon 35, c.4519_4520del. This sequence change results in an amino acid frameshift and creates a premature stop codon 37 amino acids downstream of the change, p.Gln1507Alafs*37. This sequence change is predicted to result in an abnormal transcript, which may be degraded, or may lead to the production of a truncated POLE protein with potentially abnormal function. This deletion does not appear to have been previously described in individuals with POLE-related disorders and has also not been described in population databases such as ExAC and gnomAD. The functional significance of this sequence change is not known at present and its contribution to this individual's disease phenotype cannot definitively be determined.
