Uncertain significance for Hereditary sensory neuropathy-deafness-dementia syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001130823.3(DNMT1):c.1280+2T>C, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 17 of the DNMT1 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in DNMT1 cause disease. This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with DNMT1-related conditions. ClinVar contains an entry for this variant (Variation ID: 2440976). In summary, the available evidence is currently insufficient to determine the role of this variant in disease.
