NM_000128.4(F11):c.1837G>A (p.Glu613Lys) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: F11 c.1837G>A (p.Glu613Lys) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant was absent in 251462 control chromosomes. c.1837G>A has been observed in the heterozygous state in at least 1 individual(s) affected with partial Factor XI Deficiency Disease (Rimoldi_2018). These data do not allow any conclusion about variant significance. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in 30%-50% of normal activity in an in vitro COS1 cell line assay (example, Rimoldi_2018). This variant was also reported heterozygous in at least 1 individual in the UK Biobank study (example, Stefanucci_2023). The following publications have been ascertained in the context of this evaluation (PMID: 37647632, 29178608). ClinVar contains an entry for this variant (Variation ID: 2440180). Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
