Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5854A>C (p.Asn1952His), citing Ambry Variant Classification Scheme 2023: The p.N1952H variant (also known as c.5854A>C), located in coding exon 43 of the POLE gene, results from an A to C substitution at nucleotide position 5854. The asparagine at codon 1952 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
