NM_000257.4(MYH7):c.481G>A (p.Ala161Thr) was classified as Uncertain Significance for Cardiomyopathy by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the MYH7 gene (transcript NM_000257.4) at coding-DNA position 481, where G is replaced by A; at the protein level this means replaces alanine at residue 161 with threonine — a missense variant. Submitter rationale: This missense variant replaces alanine with threonine at codon 161 of the MYH7 protein. Computational prediction tools indicate that this variant has a deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in heterozygous state in an individual affected with left ventricular noncompaction (PMID: 38380180). It has also been reported in homozygous state in an individual affected with left ventricular noncompaction (PMID: 34752814); of note this individual's homozygous sibling and heterozygous parent were clinically unaffected. This variant has been identified in 3/251482 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
