This variant lies in the GUSB gene (transcript NM_000181.4) at coding-DNA position 1192, where C is replaced by T; at the protein level this means replaces arginine at residue 398 with cysteine — a missense variant. Submitter rationale: Variant summary: GUSB c.1192C>T (p.Arg398Cys) results in a non-conservative amino acid change located in the Glycoside hydrolase family 2, catalytic domain (IPR006103) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 3.2e-05 in 251438 control chromosomes. c.1192C>T has been observed in multiple individuals affected with Mucopolysaccharidosis Type VII (Sly Syndrome)(Zhou_2021, Sun_2018, Xiang_2024). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
