This variant lies in the TAF1 gene (transcript NM_004606.5) at coding-DNA position 551, where G is replaced by T; at the protein level this means replaces serine at residue 184 with isoleucine — a missense variant. Submitter rationale: Variant summary: TAF1 c.611G>T (p.Ser204Ile) results in a non-conservative amino acid change in the encoded protein sequence. Two of four in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 183387 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.611G>T in individuals affected with Mental Retardation, X-Linked, Syndromic 33 and no experimental evidence demonstrating its impact on protein function have been reported.
