The variant allele was found at a frequency of 4e-06 in 251472 control chromosomes (gnomAD). c.4748G>A has been reported in the literature in at least four compound heterozygous individuals affected with Nonsyndromic Hearing Loss and Deafness, Type 9 (Iwasa_2013, Zhang_2013, Qiu_2019, Iwasa_2019). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as likely pathogenic.
