Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000008.10:g.(?_126369441)_(126370080_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 6-7 of the NSMCE2 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to disrupt the C-terminus of the protein. This variant has not been reported in the literature in individuals affected with NSMCE2-related conditions. This variant disrupts a region of the NSMCE2 protein in which other variant(s) (p.Ala234Glufs*4) have been determined to be pathogenic (PMID: 25105364). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
