NC_000003.11:g.(?_167405002)_(167413530_?)del was classified as Pathogenic for Cerebral cavernous malformation 3 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): A similar copy number variant has been observed in individual(s) with cerebral cavernomas (Invitae). This variant is a gross deletion of the genomic region encompassing exon(s) 5-7 of the PDCD10 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to disrupt the C-terminus of the protein. This variant disrupts a region of the PDCD10 protein in which other variant(s) (p.Phe174Leufs*3) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
