This variant results in the deletion of part of exon 2 (c.-12+15599_17del) which includes the initiator methionine of the DNAJC5 mRNA. There are no downstream in-frame methionine residues, therefore it is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in DNAJC5 cause disease.
