Pathogenic for Kleefstra syndrome 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000009.11:g.(?_140637803)_(140638562_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 5-6 of the EHMT1 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in EHMT1 are known to be pathogenic (PMID: 16826528, 19264732). This variant has not been reported in the literature in individuals affected with EHMT1-related conditions. For these reasons, this variant has been classified as Pathogenic.
