Pathogenic for Hereditary spastic paraplegia 31 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.11:g.(?_86479060)_(86481957_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 4-5 of the REEP1 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in REEP1 are known to be pathogenic (PMID: 18321925, 18644145, 22703882). A similar copy number variant has been observed in individuals with hereditary spastic paraplegia (PMID: 24451228). For these reasons, this variant has been classified as Pathogenic.
