Pathogenic for Leber congenital amaurosis 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.10:g.(?_10041069)_(10042759_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 4-5 of the NMNAT1 gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. A similar copy number variant has been observed in individuals with Leber congenital amaurosis and/or retinitis pigmentosa (PMID: 26316326; Invitae). For these reasons, this variant has been classified as Pathogenic.
