Pathogenic for Leber congenital amaurosis 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.10:g.(?_10032132)_(10032266_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. A similar copy number variant has been observed in individual(s) with Leber congenital amaurosis (PMID: 32165824). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is a gross deletion of the genomic region encompassing exon(s) 2 of the NMNAT1 gene, which includes the initiator codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NMNAT1 are known to be pathogenic (PMID: 22842229).
