Uncertain significance for Gamma-aminobutyric acid transaminase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_8851594)_(9017290_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 6-16 of the ABAT gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. This variant has not been reported in the literature in individuals affected with ABAT-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant disrupts a region of the ABAT protein in which other variant(s) (p.Leu211Phe) have been observed in individuals with ABAT-related conditions (PMID: 25738457). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
