Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.859G>C (p.Asp287His), citing Ambry Variant Classification Scheme 2023: The p.D287H variant (also known as c.859G>C), located in coding exon 9 of the POLE gene, results from a G to C substitution at nucleotide position 859. The aspartic acid at codon 287 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
