Pathogenic for Adams-Oliver syndrome 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000009.11:g.(?_138392557)_(141016451_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the NOTCH1 gene has been identified. Loss-of-function variants in NOTCH1 are known to be pathogenic (PMID: 16025100, 21457232, 25132448, 25963545). The boundaries of this event are unknown as they extend beyond the assayed region for this gene and therefore may encompass additional genes. Isolated whole-gene deletions of NOTCH1 have not been reported in the literature. However, larger copy number events that include this gene have been reported (PMID: 19597493, 22318994, 25516202, 26820064). For these reasons, this variant has been classified as Pathogenic.
