Pathogenic for Joubert syndrome; Meckel-Gruber syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_53692306)_(53698941_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 10-12 of the RPGRIP1L gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in RPGRIP1L are known to be pathogenic (PMID: 17558409). This variant has not been reported in the literature in individuals affected with RPGRIP1L-related conditions. For these reasons, this variant has been classified as Pathogenic.
