Pathogenic for Ichthyosis linearis circumflexa — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.9:g.(?_147443608)_(147484583_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 1-16 of the SPINK5 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 16 of the SPINK5 gene. This is expected to result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with SPINK5-related conditions. Loss-of-function variants in SPINK5 are known to be pathogenic (PMID: 11511292, 11841556). For these reasons, this variant has been classified as Pathogenic.
