Pathogenic for Niemann-Pick disease, type C2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000014.8:g.(?_74951108)_(74953149_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. A similar copy number variant has been observed in individual(s) with Niemann-Pick Type C (PMID: 27271431). This variant is a gross deletion of the genomic region encompassing exon(s) 2-3 of the NPC2 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in NPC2 are known to be pathogenic (PMID: 25145893).
