Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.11:g.(?_112779835)_(112780119_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a sub-genic deletion of the genomic region encompassing exon 18 of the MERTK gene. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product. This variant has not been reported in the literature in individuals with MERTK-related conditions. This variant disrupts the C-terminus of the MERTK protein. Other variant(s) that disrupt this region (p.Trp837Glyfs*6, p.Arg844Cys) have been determined to be pathogenic (PMID: 31106028, 15111602, 28462455). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. For these reasons, this variant has been classified as Pathogenic.
