Pathogenic for Mendelian susceptibility to mycobacterial diseases due to complete IL12RB1 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000019.9:g.(?_18171912)_(18177527_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 12-15 of the IL12RB1 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in IL12RB1 are known to be pathogenic (PMID: 9603733, 12591909). This variant has not been reported in the literature in individuals affected with IL12RB1-related conditions. For these reasons, this variant has been classified as Pathogenic.
