This variant disrupts a region of the DMD protein in which other variant(s) (deletion of exons 45-46) have been determined to be pathogenic (PMID: 2063877, 15723292, 21851881, 25244321). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with DMD-related conditions. This variant results in the deletion of exons 45-46 and part of exon 47 (c.6439-102508_6772del) of the DMD gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in DMD are known to be pathogenic (PMID: 16770791, 25007885).
