Uncertain significance for Charcot-Marie-Tooth disease dominant intermediate C — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.10:g.(?_33247985)_(33248160_?)del, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with YARS-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 9 of the YARS gene. This deletion is out-of-frame, and is expected to create a premature translational stop signal and result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in YARS cause disease.
