Pathogenic for T-cell immunodeficiency, congenital alopecia, and nail dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_26861752)_(26869835_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the FOXN1 protein in which other variant(s) (p.Thr487Profs*41) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with FOXN1-related conditions. This variant results in the deletion of exon 8 and part of exon 7 (c.1166_*5384del) of the FOXN1 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
