NC_000022.10:g.(?_24143121)_(24145619_?)dup was classified as Pathogenic by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing exon(s) 4-5 of the SMARCB1 gene. While the exact position of this variant cannot be determined from the data, sub-genic copy number gains are generally in tandem (PMID: 25640679). This variant is predicted to be out-of-frame, and may result in an absent or disrupted protein product. Loss-of-function variants in SMARCB1 are known to be pathogenic (PMID: 10521299, 21208904). A similar copy number variant has been observed in individual(s) with teratoid/rhabdoid tumors (PMID: 20848638; Invitae). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic.
