NC_000005.9:g.(?_158753683)_(158753790_?)del was classified as Pathogenic for Mendelian susceptibility to mycobacterial diseases due to complete IL12B deficiency by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant is a gross deletion of the genomic region encompassing exon(s) 2 of the IL12B gene, which includes the initiator codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in IL12B are known to be pathogenic (PMID: 11753820, 23429356). This variant has not been reported in the literature in individuals affected with IL12B-related conditions.
