Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_27441393)_(28899063_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the SH2B1 gene has been identified. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SH2B1 cause disease. The boundaries of this event are unknown as they extend beyond the assayed region for this gene and therefore may encompass additional genes. Isolated whole-gene deletions of SH2B1 have not been reported in the literature. However, larger copy number events that include this gene have been reported (PMID: 19966786, 20799338, 20808231, 23554873, 23860047, 27257017, 29976977, 30208311, 30518945, 31801954, 33144682, 34015165). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
