Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.11:g.(?_65707455)_(65707616_?)dup, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing exon(s) 14 of the EYS gene. While the exact position of this variant cannot be determined from the data, sub-genic copy number gains are generally in tandem (PMID: 25640679). This variant is predicted to be out-of-frame, and may result in an absent or disrupted protein product. Loss-of-function variants in EYS are known to be pathogenic (PMID: 18836446, 20333770). A similar copy number variant has been observed in individuals with retinitis pigmentosa (PMID: 30718709, 31074760). This variant is also known as c.(2137+1_2137-1)_(2259+1_2260-1)dup. For these reasons, this variant has been classified as Pathogenic.
