This variant disrupts a region of the GRM6 protein in which other variant(s) (p.Leu668Pro) have been determined to be pathogenic (PMID: 30718709; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with GRM6-related conditions. This variant results in the deletion of exons 7 and 8, part of exons 6 and 9, and the insertion of 8 nucleotides (c.1218_2354delinsGGTGCTCC) of the GRM6 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to disrupt the C-terminus of the protein.
