Pathogenic for Tuberous sclerosis 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.9:g.(?_2089925)_(2115656_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 1-16 of the TSC2 gene, which includes the initiator codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in TSC2 are known to be pathogenic (PMID: 10205261, 17304050). A similar copy number variant has been observed in individual(s) with clinical features of tuberous sclerosis (PMID: 29932062). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic.
