Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_39738787)_(39739685_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exon 7 and part of exon 6 (c.1076_1322-3del) of the KRT14 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in KRT14 cause disease. This variant has been observed in individual(s) with autosomal dominant epidermolysis bullosa simplex (Invitae). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic.
