Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.10:g.(?_22151620)_(22208639_?)dup, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing exon(s) 12-15 of the PHEX gene. While the exact position of this variant cannot be determined from the data, sub-genic copy number gains are generally in tandem (PMID: 25640679). This variant is predicted to be out-of-frame, and may result in an absent or disrupted protein product. Loss-of-function variants in PHEX are known to be pathogenic (PMID: 9097956, 9106524, 19219621). A similar copy number variant has been observed in individual(s) with hypophosphatemia (Invitae). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic.
