Pathogenic for Neuronal ceroid lipofuscinosis 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.10:g.(?_40555072)_(40558194_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 2-5 of the PPT1 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in PPT1 are known to be pathogenic (PMID: 10679943, 21990111). This variant has not been reported in the literature in individuals affected with PPT1-related conditions. For these reasons, this variant has been classified as Pathogenic.
