This variant lies in the MYOC gene (transcript NM_000261.2) at coding-DNA position 1111, where T is replaced by C; at the protein level this means replaces tyrosine at residue 371 with histidine — a missense variant. Submitter rationale: The c.1111T>C variant in MYOC is a missense variant predicted to cause substitution of Tyrosine by Histidine at amino acid 371 (p.Tyr371His). This variant was not found in any genetic ancestry group of gnomAD (v4.1.0), meeting the ≤ 0.0001 threshold set for PM2_Supporting in a genetic ancestry group of at least 10,000 alleles. The REVEL score = 0.875, which was within the 0.773-0.931 range for PP3_Moderate, predicting a damaging effect on MYOC function. There was no functional evidence predicting a damaging or benign impact of this variant on MYOC function. Only 2 segregations had been reported for juvenile open angle glaucoma (JOAG, Teruya et al, 2008), not meeting the ≥ 3 segregations required for PP1.
