This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. Studies have shown that a similar copy number variant results in skipping of exon 3, but is expected to preserve the integrity of the reading-frame (PMID: 8034304). A similar copy number variant has been observed in individual(s) with Neurofibromatosis type 1 (PMID: 8034304, 25631097, 26189818, 31766501). This variant is a gross deletion of the genomic region encompassing exon(s) 3 of the NF1 gene. RNA analysis indicates that a similar copy number variant induces altered splicing and likely results in a shortened protein product.
