Pathogenic for Melanoma, cutaneous malignant, susceptibility to, 8; Waardenburg syndrome type 2A; Tietz syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.11:g.(?_69985874)_(69990502_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with MITF-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 1-4 of the MITF gene, which includes the initiator codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MITF are known to be pathogenic (PMID: 8659547, 20127975).
