Pathogenic for Melanoma, cutaneous malignant, susceptibility to, 8; Waardenburg syndrome type 2A; Tietz syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.11:g.(?_69985874)_(70014399_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the MITF gene has been identified. Loss-of-function variants in MITF are known to be pathogenic (PMID: 8659547, 20127975). The boundaries of this event are unknown as they extend beyond the assayed region for this gene and therefore may encompass additional genes. Isolated whole-gene deletions of MITF have not been reported in the literature. However, larger copy number events that include this gene have been reported (PMID: 17318840). For these reasons, this variant has been classified as Pathogenic.
