NC_000012.11:g.(?_121174454)_(121176447_?)del was classified as Pathogenic for Deficiency of butyryl-CoA dehydrogenase by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the ACADS protein in which other variant(s) (p.Arg272Cys) have been determined to be pathogenic (PMID: 27051597; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with ACADS-related conditions. This variant results in the deletion of exons 3-6 and part of exon 7 (c.211-335_907del) of the ACADS gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in ACADS are known to be pathogenic (PMID: 12736383, 18523805).
