Submitter rationale: Variant summary: COL9A3 c.472C>T (p.Pro158Ser) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00011 in 248562 control chromosomes, predominantly at a frequency of 0.0015 within the East Asian subpopulation in the gnomAD database. The observed variant frequency within East Asian control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for a pathogenic variant in COL9A3 causing Multiple Epiphyseal Dysplasia phenotype. To our knowledge, no occurrence of c.472C>T in individuals affected with Multiple Epiphyseal Dysplasia and no experimental evidence demonstrating its impact on protein function have been reported.
