Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_194248.3(OTOF):c.5323G>C (p.Asp1775His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 1775 of the OTOF protein (p.Asp1775His). This variant is present in population databases (rs776992000, gnomAD 0.03%). This missense change has been observed in individual(s) with deafness (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
