While this particular variant has not been reported in the literature, truncating variants in CCDC39 are known to be pathogenic (PMID: 21131972, 23255504). This sequence change deletes 2 nucleotides from exon 2 of the CCDC39 mRNA (c.163_164delAT), causing a frameshift at codon 55. This creates a premature translational stop signal (p.Met55Aspfs*8) and is expected to result in an absent or disrupted protein product.
