Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_177438.3(DICER1):c.3374G>A (p.Ser1125Asn), citing Sema4 Curation Guidelines. This variant lies in the DICER1 gene (transcript NM_177438.3) at coding-DNA position 3374, where G is replaced by A; at the protein level this means replaces serine at residue 1125 with asparagine — a missense variant. Submitter rationale: The DICER1 c.3374G>A (p.S1125N) variant has not been reported in the literature to our knowledge. It was observed in 2/282212 chromosomes across all populations, in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 242084). In silico tools suggest the impact of the variant on protein function is benign, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
