Pathogenic for Medium-chain acyl-coenzyme A dehydrogenase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000016.6(ACADM):c.136_158dup (p.Phe55fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ACADM gene (transcript NM_000016.6) at coding-DNA position 136 through coding-DNA position 158, duplicating 23 bases; at the protein level this means shifts the reading frame starting at phenylalanine residue 55, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with ACADM-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Phe55Asnfs*31) in the ACADM gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ACADM are known to be pathogenic (PMID: 16121256, 20434380).
